Here is a question about ethanol in an alcoholic beverage.?

Okay so if I have a 1L bottle of vodka at 40% alcohol - I pour 500mL into 2 separate gles. Although ethanol is very soluble in water, is it possible for there to be even a Slight difference in percentages of alcohol found in each gl? And if so, why would there be these differences?
